
Engineering of riboswitch for metabolic flux control and industrial strain development
Riboswitch, a regulatory part of an mRNA molecule that can specifically bind a metabolite and regulate gene expression, is attractive for engineering biological systems. In particular, it is of interest for dynamical control of metabolic fluxes to improve the formation of target products in industrial microorganisms. In this project, we demonstrated the successful use of lysine riboswitch and intracellular L-lysine as a signal to control the competing but essential metabolic by-pathways of lysine biosynthesis in Corynebacterium glutamicum and E. coli . Work is now focused on designing synthetic riboswitches for more broad applications in industrial biotechnology. Stichworte
