Hydrolasen für Synthese und Hydrolyse von biologisch aktiven reinen Enantiomersubstanzen (Antibiotika, Peptide) Wiss. Kontakte und Kooperationen: Dept. Biotechnology, KTH Stockholm; Inst. Org. Chem., Bulg. Acad. Sci., Sofia In diesem Vorhaben werden folgende enzymkatalysierten Prozessen mit biotechnischer Bedeutung geforscht bzw. Grundlagen dafür erarbeitet: - Hydrolysen von Penicillin G und Cephalosporinen
- Kinetisch kontrollierte Synthese von semisynthetischen Penicillinen und Cephalosporinen, Peptiden und anderen Kondensationsprodukten
- Stereoselektive Hydrolysen und Synthesen
Bei diesen enzymtechnischen Prozessen ist der Endpunkt (Gleichgewicht oder Maximum) der Reaktion neben der Anfangsgeschwindigkeit von entscheidender Bedeutung. Die Ausbeuten an den Endpunkten werden von folgenden Faktoren beeinflußt: - Struktur, Sequenz- und Stereospezifität der benutzten Enzyme (Hydrolasen)
- Art der Immobilisierung des Enzyms
- pH, Ionenstärke, Temperatur, Druck und Lösemittelzusammensetzung
Die Ergebnisse der Untersuchung dieser Faktoren erlauben optimale Enzyme, Träger für Immobilisierung bzw. Reaktionsbedingungen rational auszuwählen. Für die Optimierung der Ausbeute und für das Erzielen optischer Reinheit der Kondensationsprodukte sind die Faktoren so gewählt werden, daß die Nebenproduktbildung und Produktumwandlung bzw. das Downstream processing minimiert werden. Die Stereospezifität in der S1- S1'-, und S2'- Bindungsstellen der benutzten Hydrolasen (Penicillin Amidase, Proteinasen aus mesophilen und thermophilen Organismen) bei gleichgewichtskontrollierten Hydrolysen und kinetisch kontrollierten Synthesen wird als Funktion der Primärstruktur, Temperatur, pH, und Druck bestimmt. Dieses, kombiniert mit bioinformatischer Analyse und molekularer Modellierung, ermöglicht eine rationelle Analyse und Anwendung des stereoselektiven Potentials der benutzten Enzyme.
